草菇翻译起始因子Vv-IF4E及与其共表达转录因子的研究

Abstract:

真核翻译起始因子IF4E(initiationfactor4E)可通过与mRNA的5’帽子端结合,在蛋白质的翻译起始过程中扮演重要角色。草菇Volvariellavolvacea是一种富有商业价值的食用真菌,其生长发育与蛋白质的合成代谢密切相关。本文通过鉴定草菇编码IF4E的基因(Vv-IF4E),并根据生物信息学和实时荧光定量数据分析Vv-IF4E及相关转录因子基因表达量的变化规律。结果显示,草菇Vv-IF4E基因上游存在较多顺式作用元件,基因存在4种可变剪切体,只有一种具有翻译起始因子的保守结构域。预测开放阅读框(ORF)长度为3083bp,所编码蛋白分子量为87.1kDa,存在35个磷酸化位点。Vv-IF4E蛋白结构有异于拟南芥IF4E,但与皱木耳AuriculariadelicataIF4E相似。实时荧光定量结果表明,Vv-IF4E与转录因子YRR1、ECM22存在极强的共表达规律。
