西安市典型景观水体水质与微生物种群结构多样性

Abstract:

选取西安市区9个景观水体,在分析水体水质指标基础上,运用巢式pcr-dgge和克隆测序技术研究景观水体细菌和真菌的群落结构多样性.结果表明,9个景观水体均为劣ⅴ类水.水质指标中总氮(tn)含量最高为丰庆公园,为11.286mg/l,是曲江池的5倍;硝氮(no3-n)含量最高为革命公园,为5.614mg/l,是曲江池的5.6倍;氨氮(nh4-n)含量最高为丰庆公园,为3.442mg/l,是大明宫太液池的12倍;高锰酸盐指数(codmn)最高为丰庆公园,为38.886mg/l,是曲江池的3.2倍.巢式pcr-dgge图谱主成分分析(pca)结果显示,莲湖公园、永阳公园和大明宫太液池水体细菌种群相似度高,丰庆公园、新纪元公园、曲江池和革命公园水体细菌种群多样性相似,而兴庆湖水体细菌群落结构与其他水体差异显著(p<0.05).莲湖公园、永阳公园和大明宫太液池水体细菌种群结构主要受水体no3-n、tn和do含量的调控,其中no3-n含量影响显著;nh4-n含量对革命公园、新纪元公园和曲江池水体细菌种群影响显著,其中no3-n和codmn呈现负相关关系.codmn含量对兴庆湖水体细菌种群多样性影响显著.大明宫太液池、莲湖公园、丰庆公园、新纪元公园和兴庆湖水体真菌种类高度相似,革命公园水体真菌群落结构与其他水体存在差异,种群受到tn、no3-n和nh4-n的综合调控.no2-n含量对永阳公园水体真菌种群影响最大,同时与酸碱度(ph)和溶解氧(do)含量呈现负相关.茎点霉属(phomasp.)为革命公园水体中优势真菌.可见,西安市典型景观水体悬浮细菌和真菌群落结构特征受到不同水质指标的综合调控.表4图2参27
