M¨¦thodes : L¡¯effet de la p-PD sur la viabilit¨¦ et celui sur la prolif¨¦ration cellulaire ont ¨¦t¨¦ ¨¦valu¨¦s par la m¨¦thode d¡¯exclusion du bleu de trypan. Son effet sur l¡¯ADN des cellules P3X63Ag8.653 a ¨¦t¨¦ analys¨¦ apr¨¨s une extraction d¡¯ADN et une ¨¦lectrophor¨¨se sur gel d¡¯agarose puis confirm¨¦ par la technique TUNEL en cytom¨¦trie en flux. R¨¦sultats : Un blocage de la prolif¨¦ration avec diminution de la viabilit¨¦ cellulaire ont ¨¦t¨¦ observ¨¦s. La fragmentation de l¡¯ADN dose-d¨¦pendante t¨¦moignant de l¡¯effet apoptotique a ¨¦t¨¦ d¨¦tect¨¦e sur gel d¡¯agarose et confirm¨¦e par cytom¨¦trie en flux (technique TUNEL). Conclusion : Les r¨¦sultats obtenus montrent que la p-PD induit in vitro un blocage de la prolif¨¦ration cellulaire et une apoptose dose-d¨¦pendante des cellules tumorales murines. Aims: The mechanism of toxicity and genotoxicity of p-Phenylenediamine (p-PD), a component of human permanent hair dye and suspected to be a carcinogen, on the growth of murine tumor cells (P3X63Ag8.653) was investigated. Methods: The effect of p-PD on cell viability and proliferation was evaluated by the trypan blue exclusion method. The effect on the P3X63Ag8.653 cell DNA was analyzed by gel electrophoresis and confirmed by flow cytometry analysis (TUNEL technique). Results: A dose-dependent DNA fragmentation analyzed by gel electrophoresis shows a strong correlation with the flow cytometry analysis (TUNEL technique), suggesting that the effect of p-PD on overall viability and proliferation is mediated by DNA damage and an increase in apoptosis.
