The cardiotoxicity and multimechanismdrug resistance shown by using this drug keep researchers around the world working onthe development of Doxorubicin derivatives which are more potent, less toxic and active withDoxorubicin resistant-cancer cell. This review covers the work since Doxorubicin has been found,followed by the search of more potent synthetic derivatives. Then site-specific drugs were investigatedby antibody-directed enzyme prodrug therapy (ADEPT) and antibody-drug conjugation (ADC). Atleast, the new drug delivery methods were investigated by using liposome-encapsulated deliverydrugs.
