目的:探讨DNA损伤剂甲磺酸甲酯(MMS)对SD大鼠心血管系统的影响。方法:采用腹腔注射方式给予SD大鼠甲基磺酸甲酯(25 mg/kg或60mg/kg)。经1天或3天后处死大鼠,并检测以下指标:外周血细胞数,血压,心率,血管壁超微结构,胸主动脉张力以及血小板聚集情况。结果:研究显示,与对照组相比,在两个剂量组中MMS均能够显著减少外周血白细胞数量;但是心率和血压则不受影响。此外,MMS对胸主动脉张力没有影响,电镜分析也发现其对胸主动脉血管壁超微结构没有损伤。最后,对对照组和MMS处理组进行ADP诱导的血小板聚集反应分析发现,两组之间差异无统计学显著性。结论:综上所述,不同于MMS的遗传毒性特征,其对SD大鼠的心血管系统无明显毒性。
Objective: To investigate the effects of DNA damage agent methyl mesylate (MMS) on cardiovascular system in Sprague-Dawley (SD) rats. Methods: SD rats were intraperitoneally injected with MMS (25 or 60 mg/kg). One day or 3 days later, rats were sacrificed and the following parameters were measured and examined: peripheral blood cell count, blood pressure, heart rate, ultrastructure of the vascular wall, thoracic aorta tension, and platelet aggregation. Results: It was found that MMS could significantly decrease the white blood cell counts at both doses compared to controls. On the other hand, heart rate and blood pressure were not affected. In addition, MMS had no effect on the tension in thoracic aorti, and electromicroscopy analysis also revealed no significant damage to the thoracic aortic ultrastructure. Finally, ADP-induced platelet aggregation was evaluated in both MMS-treated and control rats, and the results showed no significant difference between the two groups. Conclusion: Taken together, it is suggested that MMS does not have any significant effect on the cardiovascular system in the rat model.
