Optimization of Aqueous Enzymatic Extraction Technology of Hemp Seed Oil by Response Surface Methodology %A 所怡祯 %A 张玉红 %J Botanical Research %P 339-348 %@ 2168-5673 %D 2024 %I Hans Publishing %R 10.12677/br.2024.133036 %X 本文以火麻仁为原料,采用水酶法提取火麻仁油,在确定碱性蛋白酶为最佳提取酶的条件下。在单因素实验(酶解时间、酶解温度、pH值和料液比)的基础上,利用Design-Expert V8.0.6.1试验设计软件的 Box-Behnken Design多因素试验方法,进行响应面法分析,优化水酶法提取火麻仁油的工艺条件。结果表明:在碱性蛋白酶的作用下,料液比为1:6时,酶解温度52?C,酶解时间2 h,酶解pH为9.2的条件下,火麻仁油的得率理论值可达36.30%。按照最佳提取工艺条件获得火麻仁油的利率为36.22% ± 0.05,经方差分析实际值与理论值差异不显著(P > 0.05),表明该最优提取工艺稳定可靠,能用于火麻仁油的提取,这为火麻及火麻仁的进一步开发利用提供实验基础和数据支撑。
In this paper, seed oil of hemp was extracted by aqueous enzyme method, and alkaline protease was determined as the best extraction enzyme. On the basis of single factor experiments (enzymolysis time, enzymolysis temperature, pH value and solid-liquid ratio), response surface analysis was carried out using Box-Behnken Design multi-factor test method of Design-Expert V8.0.6.1 test Design software, and the technological conditions of aqueous enzymatic extraction of hemp seed oil were optimized. The results showed that under the condition of solid to liquid ratio 1:6, hydrolysis temperature 52?C, enzymolysis time 2 h and pH 9.2, the theoretical yield of hemp seed oil could reach 36.30%. According to the optimal extraction conditions, the yield of hemp seed oil was 36.22% ± 0.05, and there was no significant difference between the actual and the theoretical value by variance analysis (P > 0.05), indicating that the optimal extraction process was stable and reliable, and could be used for the extraction of hemp seed oil, which provided experimental basis and data support for the further development and utilization of hemp seed. %K 火麻,种子油,水酶法,响应面法,提取工艺
