Effects of soybean isoflavone on mRNA expression of ER-alpha,NGF,IL-2 and AR in cerebellum of ovariectomized rat

Abstract:

To observe the direct accommodation and effect of different dosage soybean isoflavone on the cerebellum of ovariectomized rat, the expression and distribution of estrogen receptor alpha (ERα), nerve growth factor (NGF), interleukin-2 (IL-2) and androgen receptor (AR) mRNA in the cerebellum was studied by establishing the model of ovariectomized rat and using in situ hybridization technique. The results indicate that the hybridizational signals of ERα, NGF, IL-2 and AR were mainly distributed in stratum Purkinje cell of the cerebellar cortex, nucleus fastigii, nucleus interpositus and nucleus dentatus of cerebellum, meanwhile their weak signals could also be found in the molecular and granular layer of the cerebellar cortex. In addition, the positive production mostly located in nucleus of the positive cells, and small amounts located in cytoplasm, cytomembrane or dendrite. The general tendency of the expression of ERα, NGF, IL-2 and AR positive production in the cerebellar cortex and nuclei of OVX rat decreased significantly in ovariectomized rats, while the intensity and quantity of the hybridization signals ascended with the supplement of soybean isoflavone. In the OVX group treated with soybean isoflavone of high dosage (OVX+H), the intensity and quantity of these hybridization signals could basically reach the level of those in the sham group. In short, these findings indicated that soybean isoflavone could upregulate the expression of ERα, NGF, IL-2, AR in genic levels with dose-dependent, and the similarity of their changing tendency implied that they were correlated and cooperated during the course of effect of soybean isoflavone on cerebellum.
