丹参素对高脂血症大鼠脂代谢调节机制研究

Abstract:

目的:探讨丹参素对高脂血症大鼠脂代谢的影响.方法:选用清洁级雄性SD大鼠60只,随机选取12只为基础对照组,饲以普通饲料,其余大鼠饲以高脂饲料,造模成功后随机分为高脂对照组和注射低、中、高(10,20,40mg·kg-1·d-1)剂量丹参素溶液(溶解于生理盐水)的试验组,2个对照组腹腔注射等体积生理盐水,每天腹腔注射1次,连续30d.检测血清TG,TC,HDL-C及LDL-C含量及肝脏ACC1,FAS,HMGR,CPT-I的mRNA表达水平.结果:注射丹参素后,大鼠血清中TG,TC,LDL-C含量降低,肝脏FAS和HMGR的mRNA表达水平显著降低.结论:丹参素通过降低TC含量,抑制LDL-C水平,及时清扫多余的胆固醇,有效调节高脂血症大鼠血脂水平;通过降低FASmRNA表达减少脂肪酸的合成,抑制HMGRmRNA表达来减少内源性胆固醇的合成.
