大豆HB基因GmSbh1的分离与生物信息学分析

Abstract:

从大豆叶片中分离到GmSbh1基因cDNA的完整开放阅读框(ORF)序列,对其编码的氨基酸序列、蛋白质一级、二级结构及互作蛋白等进行了生物信息学分析。结果表明:在GmSbh1基因cDNA序列的第156~1293bp处有一个完整的ORF,编码379个氨基酸;GmSbh1具有同源异型盒(homeoboxbox,HB)基因家族典型的KNOX1、KNOX2、ELK和Homeodomain(HD)结构域;检索到18条与GmSbh1核酸序列一致性大于80%的序列;GmSBH1为亲水性蛋白,不具有信号肽,分子量为42.37kDa,其中丝氨酸(Ser)、天冬氨酸(Asn)和亮氨酸(Leu)分别为占10%、9.8%和8.4%,可能发生磷酸化的位点分别在Ser(13个)、酪氨酸(Tyr)(2个)和苏氨酸(Thr)(1个)上;二级结构预测结果显示,GmSBH1序列存在α-螺旋(占39.31%)、β-转角(占5.28%)、延伸链(占8.44%)和无规则卷曲(占46.97%),不存在跨膜螺旋;蛋白互作预测表明,GmSBH1与拟南芥中的RPL、BEL1、AT4G32980.1-P、BLH2和BLH3等蛋白的互作程度较高,其中与BEL1共表达的几率较高。
