不同品种大豆叶片表皮蜡质提取后的扫描电镜观察

Abstract:

以抗线11和黑农48两个大豆品种的成熟叶片为材料,用氯仿和正己烷两种溶剂,采用常温法和加热法提取大豆叶片表皮蜡质,对脱蜡后的叶片进行扫描电镜观察。结果显示加热法效果好于常温法。采用常温法,两种溶剂都不能将大豆叶片表皮蜡质提取干净,仍有少量絮状或颗粒状蜡质残留,但氯仿的提取效果好于正己烷;采用加热法,两种溶剂不同处理时间均能将叶片表皮蜡质完全提取干净,处理时间对蜡质的提取结果有明显影响,加热30s时叶片表皮细胞均发生不同程度的变形。通过扫描电镜观察得出以氯仿为溶剂,加热60℃、处理时间15s为大豆叶片简单可行的脱蜡方法。
