Abstract:

Interleukin1β(IL-1β),thefirstinterleukintobecharacterized,playsakeyroleinregulatingtheimmuneresponse.Inthisstudy,wedeterminedthecDNAandgenomicDNAsequencesoftheIL-1βgenefromthelargeyellowcroaker,Larimichthyscrocea.PhylogeneticanalysisindicatedthattheIL-1β(LcIL-1β)genewasmostcloselyrelatedtothatofEuropeanseabass(Dicentrarchuslabrax),sharing67.8%aminoacididentity.Inhealthylargeyellowcroaker,LcIL-1βtranscriptionwasdetectedinalltestedtissues,withthehighestlevelfoundintheheadkidney.UponVibrioalginolyticusinfection,LcIL-1βtranscriptioninalltestedtissueswassignificantlyupregulated.IntraperitonealinjectionofrecombinantLcIL-1β(rLcIL-1β)improvedthesurvivalrateandreducedthetissuebacterialloadafterV.alginolyticusinfection.Inaddition,rLcIL-1βinducedmonocytes/macrophages(MO/MΦ)chemotaxisandincreasedphagocytosisandbactericidalactivityinvitro.TheseresultssuggestthatLcIL-1βplaysanimportantroleinthelargeyellowcroakerimmuneresponseagainstV.alginolyticus.
