氧化苦参碱在大鼠肠道的吸收与代谢机制

Abstract:

为系统研究氧化苦参碱(oxymatrine,omt)在肠道的吸收与代谢特征,本研究采用大鼠在体肠灌流模型,应用超高效液相色谱(ultraperformanceliquidchromatography,uplc)和高效液相色谱-电喷雾-四级杆飞行时间串联质谱[hplc-esi(+)-q-tof-ms]联用技术,对omt(100、200和400μmol·l-1)灌流液中的原型药和代谢物进行定量和定性分析。结果发现,omt的表观渗透系数和吸收率与其浓度无关(p>0.05),即omt在肠道是被动吸收;吸收量大小顺序:十二指肠>空肠>回肠>结肠(p<0.05);omt在十二指肠和空肠有两个代谢物,其中苦参碱(matrine,mt)是主要的活性代谢物。
