阿特拉津对大鼠多巴胺神经元凋亡相关因子表达的影响

Abstract:

?目的:探讨阿特拉津对多巴胺神经元凋亡相关因子表达的影响。方法:将7~8月龄的sd雄性大鼠40只随机分为4组,即对照组和阿特拉津低(10mg/kg)、中(50mg/kg)、高(100mg/kg)剂量组,每组10只,连续灌胃90d后处死,收集中脑黑质部分,通过透射电镜观察神经元细胞的凋亡情况,分别采用real-timepcr和westernblot检测凋亡相关基因bax、bcl-2、caspase-3mrna和蛋白的表达情况。结果:与对照组相比,随阿特拉津染毒剂量增加,电镜下神经元细胞线粒体出现肿胀、空泡以及线粒体自噬等现象;baxmrna及蛋白的表达随剂量增加而增加(p均<0.05);caspase-3的mrna在低剂量下表达增高(p均<0.05),但在中、高剂量下表达下降(p均<0.05),其蛋白的表达量随剂量的增加而增加(p均<0.05);但bcl-2mrna及蛋白的表达随剂量增加均呈下降趋势,差异均有统计学意义(p均<0.05)。结论:阿特拉津可通过调节线粒体凋亡相关因子的表达促进多巴胺神经元凋亡。
