生物信息学分析miR-203与VEGFA相互作用在胎盘发育中的意义 Bioinformatics reveals interaction of miR-203 and VEGFA in placentation

Abstract:

目的:通过比较正常发育情况下胎盘的基因表达谱及先兆子痫(PE)发生条件下的胎盘基因表达谱,从中找到在两者中具有共性的特征基因。方法:从Gene Expression Omnibus数据库中下载不同胎龄的正常人胎盘(GSE9984)和先兆子痫人胎盘微阵列数据(GSE6573)的基因表达谱。差异表达基因(DEGs)分析由GEO2R进行。基因组富集分析(GSEA)使用GSEA v3.0软件进行。TargetScanHuman数据库、miRTarBase数据库及mirRDB数据库用于分析可靠的miR-203作用靶基因。MiR-203与靶标基因的互作网络可视化由Cytoscape构建。结果:比较两组基因表达谱后得到共同基因10个,包括LNPEP、AKNA、SLC9A7、VEGFA、FUT9、STRBP、TMEM144、LPAR6、ERCC6L、TRAP1。通过将表达谱中比较后得到的共同基因与miR-203的靶基因进行比对,发现miR-203与VEGFA之间的作用均存在于PE发生与正常胎盘发育中。结论:MiR-203与VEGFA之间的作用可能在PE发生发展与正常胎盘发育中起着重要的作用
