Abstract:

garlic (allium sativum l.) is a very important and widely cultivated crop, which is known for its culinary and pharmacological use. until present, garlic breeding has been limited to clonal selection or mutants spontaneous or induced, because most of the germplasm of garlic is sexually sterile. however, it becomes increasingly feasible nowadays than the development tissue culture for garlic regeneration. therefore, in this study we established an in vitro regeneration protocol for this species using leaf and root segments. the effects of auxin and cytokinin level and the explants position of the leaf and root segments were studied. callus induction on apical root segments was significantly higher compared to callus induction on basal segments. callus were transferred to shoot regeneration medium consisting of ms medium supplemented with ms (t1), 0.01 mg·l-1 2,4-d and 5 mg·l-1 kin (t2), and 0.01 mg·l-1 2,4-d with 5 mg·l-1 ba (t3). shoot regeneration from callus was favored on apical root segments growing on ms medium containing 2 mg·l-1 picloran with 1 mg·l-1 2ip for 3 months. organogenesis occurred as shoot formation on ms medium containing 0.01 mg·l-1 2.4-d and 5 mg·l-1 of kin.
