1株侵染白三叶草的苜蓿花叶病毒全基因组序列分析及其寄主生物学研究

Abstract:

从杭州下沙公园种植的白三叶草中分离到1株苜蓿花叶病毒(Alfalfamosaicvirus,AMV)进行全基因组序列分析与寄主生物学研究。通过双链RNA(Double-strandedRNA,dsRNA)提取和改进的单引物扩增技术对AMV-HZ进行全基因组克隆,并测序获得了病毒的全基因序列(GenBank登录号:RNA1,HQ316635;RNA2,HQ316636;RNA3,HQ316637);对AMV的外壳蛋白(coatprotein,CP)序列进行了进化树分析,探讨了AMV病毒家族亲缘关系;设计了通用探针对AMV侵染进行了northernblot检测验证。AMV在心叶烟、本生烟、苋色藜、朝天椒上能形成侵染症状。为AMV的全基因组序列获得、分子检测和寄主生物学的研究提供了基础研究方法。
